Description
- Recognizes human APOE
- APOE (apolipoprotein E) is a plasma protein implicated in lipid metabolism
- Produced predominantly by hepatocytes, macrophages and neural cells
- Major extracellular lipid carrier in CNS
- Plays key role in neuronal protection/repair after injury
- Polymorphic with three major alleles (APOE2, APOE3 and APOE4)
- APOE2: plays protective role against Alzheimer's and heart disease
- APOE4: confers higher risk for atherosclerosis and Alzheimer's disease
Process:
- Antibody specific for APOE pre-coated onto microwells
- APOE protein in samples captured by coated antibody after incubation
- After extensive washing, another antibody specific for APOE is added to detect captured APOE protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
