Description
- Recognizes human CCL4
- CCL4 (C-C chemokine ligand 4, MIP-1β) plays key role in inflammation and immune regulation
- CCL4 produced by: immune cells (monocytes, B and T cells), fibroblasts, endothelial and epithelial cells
- Target cells include immature dendritic cells, which express cognate receptor CCR5
- Increased CCL4 expression reported in monocytes and blood of elderly individuals
- Serum concentrations found higher in patients with head and neck squamous cell carcinoma compared with controls
Process:
- Antibody specific for CCL4 pre-coated onto microwells
- CCL4 protein in samples captured by coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for human CCL4 is added to detect captured human CCL4 protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
