Description
- Recognizes human MCAM
- MCAM (melanoma cell adhesion molecule, CD146, MUC18) identified as biomarker of melanoma progression is a transmembrane glycoprotein belonging to immunoglobulin superfamily
- Consists of five immunoglobulin domains, transmembrane domain and cytoplasmic region
- CD146 primarily expressed by vascular endothelium and smooth muscle
- Key adhesion protein in vascular endothelial cell activity and angiogenesis
- Marker of circulating endothelium cells
Process:
- Antibody specific for MCAM pre-coated onto microwells
- MCAM protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for MCAM is added to detect captured MCAM protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
