Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous IL22

  • Recognizes human IL22
  • IL22 (Interleukin 22) is a member of human Type I interferon family, includes IL10
    • IL22 has potential to interact with IL10 because it binds to IL-10R2c chain with IL22R1 in its receptor complex
      • Bindings can be blocked ny soluble receptor
    • IL22 expressed in immune cells
    • Can contribute to immune disease through stimulation of inflammatory responses, S100s and defensins
    • Promotes hepatocyte survival in liver and epithelial cells in lung and gut similar to IL10

Process:

  • Antibody specific for IL22 pre-coated onto microwells
  • IL22 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for IL22 is added to detect captured IL22 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
