Description
Rapidly determine PYRase activity is streptococci and Citrobacter spp. with Thermo Scientific™ Oxoid™ Biochemical Identification System (O.B.I.S.) PYR. This rapid colorimetric test delivers simple and rapid results for the determination of PYRase activity in streptococci and Citrobacter spp, offering a rapid diagnostic alternative to time-consuming culture methods. In addition, the same enzymatic activity may be used to aid differentiation of Salmonella spp. from Citrobacter spp and other Enterobacteriaceae1,2.

The O.B.I.S. - PYR test uses Test Cards impregnated with L-pyroglutamic acid 7-amino-4-methyl-coumarin (7AMC) and dimethylaminocinnamaldehyde for the detection of PYRase activity. The enzymatic hydrolysis of this substrate by enterococci, Group A streptococci and Citrobacter spp. produces a purple color following the addition of the Developing Solution3.
We have developed a new system for PYRase testing4 using a non-carcinogenic substrate in response to health concerns associated with the use of β-naphthylamide (a potent carcinogen)5. The PYRase area on the O.B.I.S. PYR test is impregnated with L-pyroglutamic acid 7-amino-4-methylcoumarin (7AMC)4. Dimethylaminocinnamaldehyde is used as a color development reagent. The enzymatic hydrolysis of the substrate produces a purple color on addition of the O.B.I.S. PYR Developing Solution4.
