Description
- Recognizes human CRP
- C-reactive protein (CRP) is an acute-phase serum protein predominantely synthesizes by liver in response to interleukin-6 during inflammation
- Ability to react with C polysaccharide of Streptococcus pneumoniae
- Annular, pentameric protein belongs to pentraxin family of proteins
- Displays functions associated with host defense: promotes agglutination, bacterial capsular swelling, phagocytosis and complement fixation through calcium-dependent binding to phosphorylcholine
- Used as inflammation marker
Process:
- Antibody specific for CRP pre-coated onto microwells
- CRP protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for CRP is added to detect captured CRP protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
