Description
- Recognizes human IDH1
- IDH1 (PICD, IDP) belongs to isocitrate and isopropylmalate dehydrogenases family
- IDH1 may be tumor suppressor, when mutationally inactivated, contributes to tumorigenesis in part through induction HIF-1 pathway
- Common feature of major subset of primary human brain cancers
- IDH1 mutation heterozygotic, functions as dimer
Process:
- Antibody specific for IDH1 pre-coated onto microwells
- IDH1 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for IDH1 is added to detect captured IDH1 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
